Anil-Babesia rodhaini Monoclonal Antibodies: Effect against Babesia rodhaini and Cross-reactivity with Human Starin of Babesia microti OAK

Anti-Babesia rodhaini monoclonal antibodies (mAb), namely: 1-E7, 2-H2 and 3-B8, significantly suppressed the development of high parasitemia in BALB/c mice infected with B. rodhaini and all mAb-treated mice survived the infection. While, only monoclonal antibody 3-B8 showed some inhibitory effect against Babesia microti, with mice showing high parasitemia of 18.04 ± 2.69 %, at 9 days post-exposure. Westernblot analysis of B. rodhaini and B. microti parasite extract reacted with anti-B. rodhaini monoclonal antibodies showed cross-reactive bands of molecular weights 62 and 55 kilodaltons. Antigenic characterization of Brazilian bovine viral diarrhea virus isolates by monoclonal antibodies and cross-neutralization BJMBR

Nineteen Brazilian isolates of bovine viral diarrhea virus (BVDV) were characterized antigenically with a panel of 19 monoclonal antibodies (mAbs) (Corapi WV, Donis RO and Dubovi EJ (1990) American Journal of Veterinary Research, 55: 1388-1394). Eight isolates were further characterized by cross-neutralization using sheep monospecific antisera. Analysis of mAb binding to viral antigens by indirect immunofluorescence revealed distinct patterns of reactivity among the native viruses. Local isolates differed from the prototype Singer strain in recognition by up to 14 mAbs. Abstract Background Snakes of the genus Bothrops, popularly known as pit vipers, are responsible for most cases of snakebite in Brazil. Within this genus, Bothrops jararacussu and B. jararaca deserve special attention due to the severity of their bites and for inhabiting densely populated areas. Regarding the treatment of snakebites by Bothrops jararacussu, questions have been raised about the effectiveness of the specific bothropic antivenom in neutralizing myotoxic effects; however, there are no accurate data for humans. Hybrid molecules obtained through conjugation of monoclonal antibodies and toxins constitute an approach under exploration to generate potential agents for the treatment of cancer and other diseases. A frequently employed toxic component in the construction of such immunotoxins is ricin, a plant toxin which inhibits protein synthesis at ribosomal level and so requires to be internalized by the cell. Hemic proliferative disease in the bay mussel, Mytilus edulis, has been studied following mortalities in Washington State (USA). The disease has been reproduced in the labortory. The establishment of this experimental model led to study tumoral characteristics of the atypic hemocytes : ultrastrure, in vitro cultivation, transplantation. The infectious etiology of this tumor has been verified. It has been hypothesized to result from a viral infection since retrovirus-like particules have been observed following induction by bromodeoxyuridine. Determining the profile of antigen expression among meningococci is important for epidemiologic surveillance and vaccine development. To this end, two new mouse monoclonal antibodies (MAbs) have been derived against Neisseria meningitidis proteins (class 5). The MAbs were reactive against outer membrane antigens and were bactericidal. Glycosphingolipid antigens from Leishmania (L.) amazonensis amastigotes: Binding of anti-glycosphingolipid monoclonal antibodies in vitro and in vivo BJMBR
Specific glycosphingolipid antigens of Leishmania (L.) amazonensis amastigotes reactive with the monoclonal antibodies (MoAbs) ST-3, ST-4 and ST-5 were isolated, and their structure was partially elucidated by negative ion fast atom bombardment mass spectrometry. The glycan moieties of five antigens presented linear sequences of hexoses and N-acetylhexosamines ranging from four to six sugar residues, and the ceramide moieties were found to be composed by a sphingosine d18:1 and fatty acids 24:1 or 16:0. The use of monoclonal antibodies for specific pectic epitopes is an important tool in the study of the cell wall. Throughout the development of mucilage cells of Araucaria angustifolia (Bertol.) Kuntze, a gradient of distribution was observed in relation to the pectic de-esterification, as well as to the increase of galactan and arabinan epitope distribution, and to the reduction of arabinogalactans proteins (AGPs) epitope at maturity. AGP and methyl-esterified homogalacturonan (HGA) were present in the mucilage. Galactans and arabinans were also observed in the mucilage, though with weak labelling. Two hybridomas secreting monoclonal antibodies (MAbs) that react with a lipoprotein (LipL32) of the outer membrane of pathogenic Leptospira were obtained. For hybridoma production, spleen cells from BALB/c mice imunized with recombinant LipL32 (rLipL32) were fused to SP2/O-Ag14 cells, selected in HAT medium and screened in an indirect ELISA. One MAb produced was of the IgG2b isotype and the other was an IgM. The protozoan Bonamia ostreae (Ascetospora), a paasite of the flat oyster Ostrea edulis, was purified by differential and isopycnic centrifugations. Mice of the strain Balb/c were immunized with purified parasites and hybridomas were prepared by fusion of immunized mouse splenocytes with the mouse myeloma cell line P3-X63-Ag8-653. From the fusion, 12 clones were isolated and saved. The resulting antibodies were characterized with a solid phase radioimmunoassay (RIA). Monoclonal antibodies against sporangia and spores of Marteilia sp. (Protozoa: Ascetospora) ArchiMer

Digestive glands of mussels Mytilus edulis from Brittany, France, infected with Marteilia sp. (Ascetospora) were used to purify the parasite. A modification of a previously used purification protocol increased purification efficiency, permitting sporangial primordia and sporangia of Marteilia sp. to be obtained. Mouse (Balb/c) monoclonal antibodies were generated against this parasite. From the fusion, 26 monoclonal antibodies against Marteilia sp. were obtained. Monoclonal Antibodies Prepared against Theileria annulata-infected Bovine Cells Recognize an Infection-associated Antigen Expressed by the Infected Cells OAK

A battery of monoclonal antibodies were produced in mice against Theileria annulata-infected cells. Two of these antibodies (IgM and IgG1) reacted specifically with the surface membrane of the infected cells but not with Con A-stimulated or unstimulated bovine peripheral blood lymphocytes (PBL). The antibodies could stain also clones which were derived from the cell lines tested in the present study. Similarly, they reacted with T.annulata-infected but not with uninfected sheep PBL. After removal of the parasites by treating the infected cells with the anti-theilerial drug buparvaquone, the antibodies were no more able to bind to the cells.

Production and caracterization of monoclonal antibodies for the detection of Salmonella enterica in chicken meat BJM

A panel of 13 monoclonal antibodies (MAbs) that react against outer membrane proteins of Salmonella Enteritidis was obtained. Ascaris suum allergenic components (PIII) separated by gel filtration chromatography of an adult worm extract were used to immunize BALB/c mice. Popliteal lymph node cells taken from the immunized animals were fused with SP2/O myeloma cells using polyethylene glycol (MW 1450) as fusogen. The hybridomas were cultured in HAT-containing medium and cloned at limiting dilutions. Supernatants from the growing hybrids were screened by ELISA using plates coated with PIII or the A. suum crude extract. Production and characterization of monoclonal antibodies anti fragment Fc of bovine IgG BABT

The aim of this work was to produce and characterize monoclonal antibodies anti bovine immunoglobulin G (IgG). Out of seven hybridomas, two were chosen based on the ELISA'S absorbance values and were labeled B4F11 and B3H12. These monoclonals were analyzed through Western Blot for IgG fragments obtained by proteolysis with papain, separated by electrophoresis in polyacrylamide gel electrophoresis with β-mercaptoetanol as reducing agent. This revealed that, possibly, the B4F11 was directed to a conformational antigen, and that B3H12 reacted in a specific fashion with Fc (Bovine IgG crystallizable fragment). This antibody could be used in the development of reagents to immunoassays relevant for research and diagnosis.
